The workstudy objective is to forecast shelf-life (physical stability) of an

amorphous solid dispersion (ASD) in a tablet. This will be achieved by leveraging existing

physical stability data pertaining to the ASD powder. This will involve conducting DVS experiments on several physical mixtures

ASD/excipients, as well as in the tablet form, in order to understand water sorption/desorption

behaviour of the tablet formulation. Additionally, a combination of thermodynamic calculations

combined with kinetic models will be employed to project the ASD¿s shelf-life within the tablet

under different temperature and humidity conditions. These data will be compared with

available physical stability data on the tablet. All experimental procedures related to preparation

and characterization, as well as the modelling work, will be performed within the solid-state

team at Employer Pharmaceutica NV.

- Prepare physical mixtures ASD/excipients.

- Perform DVS, XRD, exploit ssNMR characterization of the mixtures and tablets.

- Run thermodynamic calculations with PC-SAFT formalism.

Qualifications

- Bachelor in Physics, Chemistry, Pharmacy or equivalent.

- Good understanding of condensed matter (crystal vs amorphous)

- Experience with different solid-state characterization equipment (XRD, TGA, DSC,

DVS, IR)
